In the presence of 2.4-D, B5 medium was used for suspension culture of soybean root cells. The concentration of nitrate is between 20-30 mM, and the additional addition of 2 mM ammonium sulfate can promote cell growth. When NH4+ was used as the sole nitrogen source, it could not support the growth of plants. Similar results were obtained when NH4NO3 was used instead of (NH4)2SO4. However, when the concentration of the ammonium salt exceeds 2 mM, growth is inhibited instead.
Changes in the concentration of phosphate, calcium and magnesium ions in the medium have relatively little effect on plant growth. Thiamine is an essential nutrient for cell growth at concentrations up to 10 mg/l.
