Swiss scientists invented a new type of rapid breathing diagnostics. When we drink alcohol or eat foods such as garlic, there is a special smell in the breath. But breathing not only exposes the food we eat, but certain diseases also produce odors. Therefore, respiratory testing can help with clinical diagnostics, and it does not require blood to pierce the skin. Due to the complexity of the instrument, past samples require very complex preparations for analysis, and only a few volatile compounds can be reliably detected. Recently, Swiss scientists have invented a mass spectrometry method that can quickly and reliably test molecules in the breath, including large, non-volatile compounds. This method, invented by the Renato Zenobi group of Zurich ETH, is based on quadrupole time-of-flight mass spectrometry (QTOF). During this process, the molecules are charged and then separated and identified based on the molecular mass. In the QTOF instrument, the molecules are accelerated by the electric field, and the time-of-flight meter separates the molecules according to the molecular charge-to-mass ratio. The time they fly to the detector depends on the mass. Eventually the instrument will acquire the spectrum and thus identify the original molecular species. The key to the Zenobi method is the way the sample is placed in the instrument. In general, the sample needs to be extracted first and then atomized in an electric field. But the Zebobi team used direct droplet extraction. During the process of entering the mass spectrometer, the droplets gradually lose their solvent, leaving only charged molecules. This allows the analysis to last a long time and allows for the analysis of large samples. Samples do not require prior preparation. Most importantly, these breath sample droplets contain large, non-volatile molecules compared to conventional methods, which allows more molecules to be detected.
